转化生长因子β促进面突外胚间充质干细胞向平滑肌细胞分化

摘要: 目的 探讨转化生长因子β(TGF-β)对面突外胚间充质干细胞向平滑肌细胞分化的影响。方法  60 pmol/LTGF-β作用于面突外胚间充质干细胞,分别于1 d、2 d后进行抗α平滑肌肌动蛋白(α-SMA)单克隆抗体免 疫组化图象分析检测,2 d后定量RT-PCR检测α-SMA mRNA的表达。结果 免疫组化显示,TGF-β诱导组表达α- SMA的细胞数约为95%,而未加分化抑制剂的自然分化组表达α-SMA的细胞数约为65%。图象分析显示,1 d、2 d TGF-β诱导组抗α-SMA着色平均灰度值小于未加分化抑制剂的自然分化组。定量RT-PCR显示TGF-β诱导组α-SMA mRNA量大于自然分化组。有分化抑制剂的对照组细胞不表达α-SMA。结论 TGF-β诱导组细胞表达α-SMA强于 自然分化组。TGF-β可以促进外胚间充质干细胞向平滑肌细胞分化。

Abstract:

Objective To investigate the effect of transforming growth factorβ(TGF-β)on ectomesenchymal stem cells differen- tiating to smooth muscle cells.Methods 60 pmol/L TGF-βwas added to the ectomesenchymal stem cells of embryonic facial processes. Immunohistochemistry assay and image analysis were used to value the expression extent ofαsmooth muscle actin (α- SMA) and quantitative RT-PCRwas used to value the quantity ofα-SMA.Results 2 days later, about 95% cells inTGF-βgroup and 65% cells in control groupwithout differentiation inhibitor expressedα-SMA. Expression ofα-SMA inTGF-βgroupwas stron- ger than that of control group after one and two days. Quantitative RT-PCR showed the quantity ofα-SMAmRNA in treated group cells was more than that of in control group.Conclusion Quantity ofα-SMA in TGF-βgroup is more than that of spontaneous differentiation group. TGF-βhas positive effect on ectomesenchymal stem cells differentiating to smooth muscle cells.
