人端粒酶逆转录酶与bcl-2在成釉细胞瘤中的表达

摘要:

目的 研究成釉细胞瘤(AB)中人端粒酶逆转录酶(hTERT)和bcl-2的表达。方法 采用原位杂交或免疫组化方法对AB(分原发、复发和恶变组)中hTERTmRNA和bcl-2进行检测,并与正常口腔黏膜作对照。结果  hTERTmRNA阳性表达率在AB中为94·4%,正常口腔黏膜中为14·3%(P<0·001);bcl-2蛋白阳性率在AB中为 88·0%,正常口腔黏膜中为44·4%(P<0·001)。从表达趋势看伴随着AB的复发与恶变,hTERTmRNA与bcl-2表达均增强(P<0·05)。结论 端粒酶和bcl-2可作为评价AB预后的有效指标。端粒酶的活化和bcl-2表达的增强在 AB的发生、发展中起着重要作用。

Abstract:

Objective To study the expression of telomerase reverse transcripatase (hTERT) and bcl-2 in ameloblastoma (AB).Methods hTERTmRNA in 54 cases of AB (primary AB 31 cases, recurrent AB 17 cases, malignant AB 4 cases) and 7 cases of oral normal mucosa was detectd by in situ hybridization, and bcl-2 by S-Pmethod.Results The expression of hTERT mRNAwas negative or weak in normal oral mucosa (14·3%), moderate or strong in AB (94·4%). There was a significant dif- ference in these two groups (P<0·001). The difference between the expressions of hTERT in primary, recurrent and malignant AB was significant (P<0·05). The positive ratio of bcl-2 in AB and normal oral mucosa was respectirehy 88·0%, 44·4%. There was a significant statistical difference in these two groups (P<0·001). hTERTmRNAwas stronger in recurred or malig- nantly transformed AB (P<0·05).Conclusion The expression of hTERTand bcl-2 is stronger in recurred ormalignantly trans- formed AB, and it could be used as an indicator of AB prognosis. Telomerase activity and bcl-2 expression play an important role in genesis and development of AB.
