溴脱氧尿嘧啶核苷体外掺入舌癌的初步研究

摘要:

目的:观察溴脱氧尿嘧啶核苷(BrdU)标记在肿瘤增殖研究中的价值。方法:用80LgPml的BrdU或碘脱氧尿嘧啶核苷(IdU)体外掺入Tca8113细胞或舌癌组织,标本连续切片与增殖细胞核抗原(PCNA)表达比较;相同孵育时间的细胞与流式细胞术(FCM)定量S期细胞数比较。结果:BrdU标记细胞核大浑圆,呈散在或姊妹细胞分布,子代细胞染色稍淡。BrdU或IdU掺入和PCNA表达之间无显著性差异(P>0105)。FCM测定的S期细胞数显著多于 BrdU掺入的细胞数(P<01001)。结论:BrdU标记可用于细胞增殖研究,其应用潜力较大。

Abstract:

Objective:The aim of this study is to evaluate of Bromodeoxyuridine (BrdU) label in the proliferation of the tongue cancer. Methods:The authors used 80LgPml BrdUoriododeoxyuridine (IdU) to label three cases of fresh tissue of tongue cancer and four bottles of Tca8113 cells in logarithmic growth period in vitro. Theywere fixed and embedded in the routine procedure, and then in serial sections. These specimenswere labeled with BrdUpolyclonal antibody (1B200), and PCNAmonoclonal antibody (1B100), stained using immunohistochemical ABC method. The authors observed the shapes and sizes of those positive cells, counted their label indexes (LI) of BrdU- or IdU-positive cells and PCNA- positive cells respectively. Four bottles of Tca8113 cells were cul- tured for 1~4 days. One bottle of the cells was every day used in the experimental study about comparing the cells labeled BrdU with those of the S phase of FCMin same cultural time.Results:The positive cells generally owned a larger round nucleus, some- times in company with sister cells. There was no difference of positive cells in these two kinds of labeling methods between the in- corporation of BrdUor IdUand the expression of proliferating cell nuclear antigen (P>0.05). The cells of S phase with FCM method were much more than thatmethod with BrdUincorporation (P<0.001).Conclusion: The method of BrdUlabeling could be used in tumor cellular proliferation studies.
