口腔鳞癌淋巴微转移中血管内皮生长因子C与诱导型一氧化氮合酶的相关性表达

Expression Related to Vascular Endothelial Growth Factor C and Induced Nitride Oxide Synthesizase in Lymph Node Mi- crometastasis of Oral Squamous Cell Carcinoma

摘要:

目的 探讨血管内皮生长因子C(VEGF-C)与诱导型一氧化氮合酶(iNOS)在口腔癌微转移中的作用及表达的相关性。方法 对47例口腔鳞癌组织标本、15例正常口腔黏膜采用RT-PCR方法检测其VEGF-C、iNOS mRNA 的表达;对10个正常淋巴结、47例口腔鳞癌患者的355个淋巴结采用免疫组织化学方法检测其细胞角蛋白(CK)的表达。结果 47例口腔鳞癌标本VEGF-C、iNOS mRNA的表达率分别为57·4%、68·1%,高于正常对照组的20·0%、 33·3%(P<0·05);27例VEGF-C mRNA阳性标本中, iNOS mRNA阳性占88·9%(24/27),二者存在明显相关性(P< 0·01);47例标本中CK阳性率为48·9%(23/47),VEGF-C、iNOS表达与CK呈正相关关系。VEGF-C、iNOS阳性组中淋巴微转移率分别为63·0%、65·6%,高于阴性组的30·0%、13·3%(P<0·05)。结论 在口腔癌微转移的机理中 VEGF-C与iNOS的表达具有相关性。

Abstract:

Objective To investigate the relationship of vascular endothelial growth factor c(VEGF-C) and induced nitride ox- ide synthesizase(iNOS) expression in lymph node micrometastasis of oral squamous cell carcinoma.Methods Samples were ob- tained from 47 cases of oral squamous cell carcinoma and 15 caseswith normal oral mucosa, VEGF-C and iNOSmRNA expression were detected by RT-PCR method. Lymph node micrometastasis of 10 normal lymph nodes and 355 lymph nodes from 47 cases of oral squamous cell carcinoma was detected with immunohistochemical reaction in cytokeratin antibody.Results The percentages in tumorswith higher expressionwere 57·4% forVEGF-C, 68·1% for iNOS(P<0·05). Theywere significantly higherthan that of normal groups. Significant positive relationshipwas found between VEGF-C and iNOS(P<0·01). The positive rate of cytoker- atin(CK) was 48·9%. Significant positive relationship was found between VEGF-C and CK, iNOS and CK(P<0·01). The ex- pression rates of CK in positive group of VEGF-C and iNOS were 63·0%,65·6% respectively,and were significant higher than negative groups.Conclusion Expression of VEGF-C and iNOS in lymph node micrometastasis of oral squamous cell carcinoma is significant related.
