摘要:

目的 采用体外微核法评价新型纳米根管充填材料nHA-PA66的基因毒性。方法 以V79细胞作为实验细胞,采用新型纳米根管充填材料nHA-PA66粉剂及混合物的浸提液作为实验组,丝裂霉素C为阳性对照组,用 MTT法检测各组的细胞毒性,以V79细胞中所形成的微核数作为标准评价材料的基因毒性。结果 实验组的MTT 值在各个时间段与阴性对照组无显著性差异(P>0·05);所形成的微核数(粉剂组:6·1±1·1/1 000;混合物组:5·7± 0·6/1 000)与阴性对照组(5·3±0·8/1 000)无显著性差异(P>0·05),而与阳性对照组(123·9±8/1 000)有显著性差异 (P<0·05)。结论 本实验显示新型纳米根管充填材料nHA-PA66无细胞毒性及基因毒性,生物相容性好。

Abstract:

Objective The micronucleus test was applied to evaluate the genotoxicity of a new nanocomplex HA-PA66 root filling materialin vitro.Methods The dulbecco′s modified eagle media(DMEM) extracts of the powder part and the mixture of the new nanomaterial were prepared separately. The V79 cellwas used as the test cell and the mitomycin C(MMC) as the positive control. The MTT assay was employed in our study to evaluate the cytotoxic effect while the number of micronucleus was used as the criteria forthe detection of genotoxocity.Results TheMTTvalues in test groups and negative groupwere not significantly dif- ferent at different times (P>0.05). The number of micronucleus in test groups (powder group: 6.1±1.1/1 000; complex group: 5.7±0.6/1 000) was similar to the negative control(5.3±0.8/1 000,P>0.05), while theywere significantly different to the positive control(123.9±8/1 000,P<0.05).Conclusion The newnanocomplexHA-PA66 root fillingmaterial showed no detectable cytotoxic and genotoxic effects in this study and was proved to be biocompatible.
