机械压力对青春期大鼠咀嚼肌细胞增殖活性影响的体外研究

Study on Mechanical Compression Regulating the Proliferation of Young Growing Rat Masticatory Myocytein vitro

摘要:

目的 建立咀嚼肌细胞的力学刺激—细胞培养模型,在细胞水平探讨机械力作用下青春期大鼠咀嚼肌细胞功能代谢的变化。方法 采用流式细胞术检测不同时段、不同力值的机械压力对体外培养的青春期大鼠咀嚼肌细胞增殖活性的影响。结果 各实验组肌细胞的DNA含量(S期百分数)均大于对照组。细胞持续受力2 h 后,实验组细胞的增殖指数(PI)较对照组高;细胞持续受力4 h后,在2 000μstrain组的细胞增殖指数达到最高 (48·9%),而4 000μstrain组的PI在所有实验组中最低(39·0%)。结论 适当的压力对咀嚼肌细胞起着促进增殖的作用;而较大的压力作用较长时间时,对体外培养咀嚼肌细胞的增殖活性却有抑制作用。

Abstract:

Objective To establish an experimental model which was the masticatory myocyte culture and force stimula- tionin vitroand study the proliferation changes of cultured masticatory myocytes caused by mechanical compression in young rat. Methods Flow cytometry (FCM) was used to examine the changes of cellular DNA content and cell cycles of cultured masticatory myocytes.Results The DNA content in experimental groupswas higherthan that in control. Afterthe compressive forcewas ap- plied for 2 hours, the proliferation index (PI) in experimental group became higher than that in control. Under a continuous pres- sure for 4 hours, the PI in 2 000μstrain group reached the maximum (48·9%) butthe PI in 4 000μstrain group reached the min- imum (39·0%).Conclusion The proliferation of masticatory myocytes from young rat increased under certain force and certain period of time, but decreased if the force applied was overloaded.
