舌鳞状细胞癌Tca8113细胞株醛脱氢酶高表达亚群细胞生物学特性的研究

摘要:

目的检测醛脱氢酶(ALDH)在舌鳞状细胞癌Tca8113细胞株中的表达,研究ALDH高表达(ALDHhig)亚群细胞的生物学特性。方法采用流式细胞仪检测Tca8113细胞株中ALDH的表达;分选ALDHhig与ALDHlow亚群细胞,体外培养观察其增殖、分化及在无血清培养基中的成球能力;利用抑制消减杂交(SSH)技术筛选ADLHhig亚群细胞高表达基因。结果舌鳞状细胞癌Tca8113细胞株中仅有1.3%的细胞高表达ADLH;分选出的ADLHhig肿瘤细胞增殖能力高于ALDHlow细胞和未分选的肿瘤细胞;随着培养时间的延长,体外培养的ALDHhig细胞比例逐渐下降至分选前水平;ALDHhig亚群细胞能够在无血清培养基中悬浮生长形成肿瘤球,且高表达干细胞相关基因。结论舌鳞状细胞癌Tca8113细胞株中有一小群细胞高表达ALDH,具有干细胞的生物学特性;ALDH可能是舌鳞状细胞癌干细胞的标志物之一。

Abstract:

Objective To detect the expression of aldehyde dehydrogenase(ALDH) in tongue squamous cell carcinoma Tca8113 cell line and observe the biological characteristics of tumor cells expressing high level of ALDH (ALDHhig). Methods The expression of ALDH in tongue squamous cell carcinoma Tca8113 cell line was detected by the flow cytometry. The ALDHhig and ALDHlow subpopulation cells were isolated and cultured in vitro respectively. The ability of differentiation, proliferation and tumorsphere formation were detected. Suppression subtractive hybridization( SSH) was employed to screen the genes overexpressed in ALDHhig subpopulation cells. Results The ALDHhig cells in tongue squamous cell carcinoma Tca8113 cell line was rarely 1.3%. Compared with ALDHlow cells and unsorted cells, ALDHhig cells demonstrated increased proliferation capacity. After culture in vitro, the percentage of ALDHhig cells decreased gradually to the level of that in unsorted tumor cells. The ALDHhig subpopulation cells could survive in serum free medium and generate tumorspheres. SSH analysis revealed these tumor cells also expressed stem cell related genes. Conclusion There is a rare subpopulation cells expressing ALDH highly, which displays characteristics similar to stem cells. ALDH may be one of markers for cancer stem cells of tongue squamous cell carcinoma Tca8113 cell line.
