Objective To explore the response of rat bone marrowmesenchymal stem cells (MSCs) and calvarial osteoblasts to mechanical strain and the consequent changes of cytoskeleton F-actin.Methods Bone marrow MSCs and calvarial osteoblasts were isolated from SD rats and culturedin vitro. Mechanical stretchwas performed on passage 3 cells at 2 000μεfor 0, 2, 6 and 12 hours using four-point bending system. The response of cells and the distribution of F-actin were observed using fluorescent staining under laser scanning confocal microscope and the morphological parameters were quantified using image analysis software Laserpix. Results Under mechanical stretch, the fluorescent staining decreased obviously at bothMSCs and osteoblasts, and F- actin filaments were rearranged and became tenuous, thinner, and abnormally distributed. The outline of nucleus became unclear and apoptotic changes were observed at some of both cells. Cellular size decreased more significantly inMSCs than in osteoblasts. Quantity analysis showed that total area of cells, total fluorescent density and green fluorescent density (F-actin) were all signifi- cantly decreased in MSCs(P<0·05 orP<0·01), and total fluorescent density, green fluorescent density and red fluorescent density (nuclei) did also in osteoblasts(P<0·05 orP<0·01).Conclusion Mechanical stretch caused extensive response on both MSCs and osteoblasts which led to the rearrangement of F-actin filament and apoptosis in some of these cells. MSCs were more sensitive to mechanical strain than osteoblasts.

Objective To observe the effect of matrix metalloproteinase-1(MMP-1) from human host on degradation of dentin organic matrix of root dentin. Methods The freshly extracted caries-free impacted teethwere selected. Teethwere cuttransverse- ly under the enamel-cementum junction into dentin sections with a thickness of about 5 mm. Then all sections with removal of ce- mentum, pulp and predentin were randomly divided into four groups. In the first group, dentin sections were demineralized with acid solution for 21 days, and then incubated withMMP-1 solution for 7 days; the second group were only treated with acid solu- tion for 21 days; the third group were only attacked byMMP-1 solution for 7 days; and the fourth group were untreated as a con- trol. Then all sectionswere dehydrated in ascending strength of alcohol, critically dried, coated with platinum, and then observed under scanning electron microscope(SEM).Results The dentin sections of root surface attacked by acid andMMP-1 showed that demineralization of dentin mineral and degradation of dentin matrix fibrae synchronously happened. The dentin matrix fibrae wasn′t degradated in the groups treatedwith acid orMMP-1.Conclusion The proteinases from human hostmay play an important role in the development of root surface caries. MMP-1 may distinctly degradate the organic matrix of demineralized dentin.

Objective To evaluate the effects of GLUMA desensitizing bonding system in desensitizing hypersensitivity of vital abutment. Methods 69 central incisors with vital pulp from 69 patients were randomly divided into A, B and C group. After tooth preparation, group A coated with primer after etching, group B coated with primer after etching cervical dentin only, and group C was control group without special treatment. The cold sensitivity of abutments was tested after coating with primer, after cementation of crown and three months later. Two fresh extracted caries and restoration-free middle incisorwere prepared in routine way. The surface of specimen 1 was etched with conditioner and the control specimen 2 was prepared without any special surface treatment. Scanning electron microscope(SEM) observation was used to examine the details on dentin surface. Results In all three times of examination, the sensitivities of two experimental groups had been significantly reduced compared with the control group. Immediately after coating, group B were more sensitive than group A (P<0.05). After cementation of crown and three months later, there was no statistically significant between group A and group B. SEMphotomicrograph showed that the smear lay- er was removed from etched dentine surface with open dentinal tubules. Conclusion Coating with GLUMA desensitizing bonding system could prevent hypersensitivity of vital abutment.

Objective To investigate morphologic changes of Angle classⅡand skeletal classⅠpatients during pendulum and straight wire appliances treatment and to analyze the mechanism of this treatment protocol. Methods 10 patients with Angle classⅡand skeletal classⅠmalocclusion were recruited. The lateral cephalometric radiographs were evaluated to determine if there were significant differences among pre-treatment(T1), after molar distalization(T2), and post-treatment(T3) variables in- cluding skeletal, dental and soft tissue relationships. Results Differences between theT1 andT2 meanswere significant for distal movement and distal angulation of the maxillary molars (P<0·01), mesial angulation of the maxillary incisors (P<0·05), mesial movement of the maxilla (P<0·01). There were no significant changes betweenT1 andT3 in respect saggital position and angulation of the maxillarymolars (P>0·05). Onlythe mesial movement of lowerfirstmolars and upper and lower jawswere sig- nificant (P<0·01) duringthewhole treatment. No upper second molar impactionwas detected aftertreatment. Conclusion The upper molars distalization could be achieved by pendulum appliance. The classⅠmolar relationship could be obtained by the dif- ferential growth of jaws and the differential movement of upper and lower molars.

Objective To study the multi-lineage potential of bone marrow mesenchymal stem cells (MSCs) derived from transgenic mice with green fluorescent protein (GFP) genein vitro. Methods A6-week-old GFP transgenic mouse was executed by dislocation of cervical vertebra, and the marrow in tibia and thighbone was washed outwith asepsis. The limited cell strains of MSCs derived from GFP transgenic mice(GFP-MSCs)were obtained with density gradient centrifugation. The passage 3 GFP-MSCs were induced to differentiate into osteoblast, adippcyte, neuron with solution of calcium induction medium, adipogenic medium and neural induction medium respectively. After being calcium-induced, the activity of alkaline phosphatase on GFP-MSCs was determined by micro-plate reader, and alizarin red staining was performed to test the formation of calcium concentration. The adipo-induced MSCs were detected with oil red O staining. Immunocytochemical stainingwas performed to detect the expression of NSE on neuron-induced MSCs.Results The ALP activity of GFP-MSCs heightened gradually alongwith being calcium-induced, and alizarin red staining showed positive. Oil red O staining of adipo-induced cells and NSE immunocytochemical staining of neu- ron-induced cells demonstrated positive. Conclusion The limited cell strain of GFP-MSCs possessesmulti-lineage potential, which can be used as an efficient tracking facility for studying the mechanism of multi-lineage potential on the MSCs.

Objective The purpose of the study is to detectthe expression ofTGF-β1 mRNA in the maxillary sutures of puberty rhesus during different periods of the loading of intermaxillary classⅢorthopedic force. Methods The animal model was estab- lished with 6 puberty female rhesus, whichwere randomly divided into experimental group(wearing classⅢtwin-blockmagnet ap- pliance, each 2 rhesus for 3 and 6 month respectively) and control group(not wearing any appliance, each 1 rhesus for 3 and 6 month respectively ). Tissue sections were obtained perpendicular to the sutures. In situ hybridization was used to the expression of TGF-β1 mRNA, and the expression intensitywasmeasured and statisticswas performed by SPSS 11.0.Results Therewere no statistic differences of cellular density between palatomaxillary suture in vertical group and pterygomaxillary suture in horizontal group, but statistic differences were found between other groups. The expression of TGF-β1 mRNA was detected in the control group, but the expression intensity was obviously increased after the load of intermaxillary classⅢorthopedic force. Statistically significant differences were found among all groups exceptthe sixmonths experimental group and control group of temporozygomatic sutures and pterygomaxillary sutures. Experimental groups were more intensive than the control group and three months group was more intensive than the six months group. Conclusion The active tissue remodeling happened in the circummaxillary sutures by the effect of classⅢintermaxillary orthopedic force. Cell proliferation, extracellularmatrix synthesis and bone formationwere ac- celerated. During the different remodeling periods, the expression intensities were different, which may be related to the different force loading manners, the different reaction of sutures tothe orthopedic force and the different biological function ofTGF-β1 in the different periods.

Objective To study the relationship of expression of mucin 1 and E-cadherin with recurrence of pleomorphic ade- noma in salivary gland, and to investigate the signal to predictthe recurrence potential of the tumor. Methods The capsule of tu- mor was observed bymicroscope. The expression ofmucin 1 and E-cadherin in 33 cases of primary adenoma, 12 cases of recurrent pleomorphic adenomas and 7 cases of malignant pleomorphic adenomas were detected by immunohistochemistry. Results There was no significant difference about the status of capsule and the positive rate of mucin 1 expression between primary and recurrent pleomorphic adenoma (P>0·05). The abnormal distribution of mucin 1 expressionwas observed in recurrent pleomorphic adeno- ma (6/8), whichwas characterized by the positive staining of the whole cytomembrane. On the other hand, positive staining of the primary pleomorphic adenomawas observed on the top of the membrane (19/21). The differencewas statistically significant (P< 0·05). The staining pattern in malignant pleomorphic adenoma was similar with the recurrent ones except higher ratio of positive expression .No significant differentwas observed among the three kind of tumors on the expression rate of E-cadherin(P>0·05). Conclusion The status of capsule didn′t have much actual usage in predicting the recurrence of pleomorphic adenoma. There was no significant relationship between the expression of E-cd and the recurrence of the tumor. The abnormal distribution of mucin 1 expression contributes to the invasiveness of the tumor and can be used as the predictive signal for recurrence of pleomorphic adenoma.

Objective The aim of this studywas to investigate howto model an accuracy 3-dimension Finite Element Analysis (3D-FEA) model. Methods Based on computed tomography (CT) scan data of a woman, a 3D finite element model of the first molar on the left was rebuilt by computer imagines process and computer aided design (CAD). Analysis of the stress distribution on a cylinder dental implant and in the bone around itwas conducted. Results The stress distribution showed extremely asymme- try in bucco-lingual section: stress concentrated on the lingual side of the mandible; stress mainlywas tensile in buccal side, and on the contrary compressive in lingual side. Conclusion The resultswere more reliable because this model more really displayed the mandible.

Objective To observe human tumor necrosis factor-α(hTNF-α) expression and secreting level of human embryo myoblasts transfected by hTNF-αgene.Methods Human embryo myoblasts were transfected with shuttle plasmid pSV23SHTNF containing hTNF-αgene by cationic liposomes DOSPER. The control group was only given equivalent liposomes except plasmid. After culturing for 24, 48, 72 and 96 hours, hTNF-αexpression level of human embryo myoblastswas observed with immunocyto- chemistry staining, and hTNF-αsecreting of human embryo myoblasts was analyzed by ELISA.Results After transfected by hT- NF-αgene for 24 , 48, 72 and 96 hours, the human embryomyoblasts displayed significant secretion of hTNF-αin the cultural su- pernatant(P<0.05), and overexprsssion in cytoplasma and cell membrane. Conclusion Transfection of hTNF-αgene to human myoblasts made myoblasts secrete high concentration of hTNF-α, implying it is feasible that transfecting muscle cells surrounding tongue carcinoma lesion with hTNF-αgene can prevent tongue carcinoma from intruding into deeper muscle tissue.
