  • Construction of Recombinant Plasmid pEGFP-BMP7 and Its Expression in Rat Bone Marrow Mesenchymal Stem Cellsin vitro

    Objective To construct recombinant plasmid pEGFP-BMP7 and determine its expression in rat bone marrow mes- enchymal stem cells (MSCs)in vitro.Methods cDNA of target gene was obtained from neonatal rat kidney by RT-PCR. After sequencing the target gene, the cDNAwas subcloned into a eukaryote plasmid pEGFP-N1 by directed cloning and then digested with two restrictive endonucleases to verify the correctiveness of the recombinant plasmid pEGFP-BMP7. Rat bone marrow MSCs were transiently transfected with the pEGFP-BMP7 and transfection efficiency of the Green Fluorescent Protein (GFP) was deter- mined. RT-PCR and immunocytochemical analysiswere also performed to detectthe expression of BMP7 in ratMSCs.Results A 1 311 bp cDNA fragment was obtained by RT-PCR and sequence analysis showed it matched perfectly with that of rat BMP7 gene except a single nucleotide change at 756 bp from T to A. Digestion of the recombinant plasmid showed two 1·3 kb and 4·7 kb fragments and their size were same as those of BMP7 and pEGFP. This indicated that BMP7 cDNAwas successfully subcloned into pEGFP. Transient transfection showed an efficiency of 33% at day 2 in rat MSCs. After transfection, transcription of BMP7 was detected in MSCs and expression of BMP7 protein was also verified.Conclusion Recombinant eukaryote plasmid pEGFP-BMP7 was successfully constructed and expressed in rat bone marrowMSCs. This procedure may provide a unique method for stimulation of callus formation in distraction osteogenesis and reconstruction of craniofacial bone defects.

    Effect of Pulsed Nd:YAG Laser Irradiation on Periodontal Ligament Fibroblast Cultures

    Objective To evaluate the effect of the pulsed Nd:YAG laser irradiation on the survival rate of human periodontal ligament fibroblast (PDLF) at different power and irradiation time settings, and to compare the morphological change of cells before and after irradiation.Methods The cells were cultured and placed into the 96-well tissue culture plates. Cells were divided into groups according to the irradiation time (10 s,20 s,40 s,60 s) and the output energy (0·5 W,1·0 W,1·5 W,2·0 W,2·5 W). After the irradiation, the cell numberwere counted and the survival rateswere calculated. The correlation between the survival rate and irradiation time as well as power output was analyzed.Results No significant reduction of survival rates were found in 10 s group, as well as in 20 s group with 0·5 W, 1.0 W setting·The increase of the power output and increase of the irradiation time caused significant reduction of cell survival rate (P<0·05). The irradiation time was more relevant to this reduction than the power output.Conclusion It indicates that the laser irradiation may cause damage to PDLF and decrease the cell survival rate. The irradiation time is more related to this effect than the power output.

    Objective To investigate the feasibility of plasmid nm23-h1 transfection on high metastatic potential adnoid cystic carcinoma(ACC-M) cell line mediated by cationic lipid.Methods ACC-M cell were implanted in the maxillofacial region in each of 40 BALB/c nude mice. After the tumor growth to 1 cm in diameter,0.1 ml Lipofctamine-nm23-h1 plasmid complexwere injected intratumorally in 10 mice, 3 days after the first injection,10 mices injected for twice, 10 mice as plamid-blank control, another 10 mice were injected 0.2 ml complex, 2,3,7days after the injection, the mice were killed and the specimen forHE and immunohistological chemistry study.Results nm23-h1 expression initiated in the tumor cells 3 days after the complex injection, 7 days later, the expression level increased accompanying with extracellular matrix increase, twice injection and multiple channel injection would gain better nm23-h1 expression than once injection and single-channel injection respectively.Conclusion Cation- ic lipid mediated nm23-h1 plamid transfecting adnoid cystic carcinoma can gain small range positive expression, but the results give little prospect for further clinical treatment in such a manner.

    Objective To investigate the expression and distribution pattern of IntegrinαVβ3(ItgαVβ3) in course of distraction osteogenesis and to quest for the function of ItgαVβ3in distraction osteogenesis (DO).Methods 44 adults New Zealand white rabbits were selected and divided randomly into 3 groups: DO, bone fracture and normal group. There were 20, 20 and 4 rabbits in each group separately. Animal models were established and animals were killed at different time points. Sections were stained by IHC method. Distribution and expression pattern of ItgαVβ3were observed by semi-quantitative technique, and the results were managed with statistic methods.Results The expression of ItgαVβ3was found both in the DO and bone fracture groups. The positive stainingwas seenmainly in vascular endothelial cells on cytomembrane and in cytoplasm. The staining intensity of group of DOwas higher than that of the bone fracture group.Conclusion ItgαVβ3plays an important role in promoting the process of DO.

    Objective To investigate the aciduric bacteria in different stages of the fissure caries in orderto determine potential roles of the microflora in the development of dental caries.Methods Plaque sampleswere taken from10 incipient carious fissures of the first permanentmolars. Plaque sampleswere also collected from sound fissures of the first permanentmolars in 10 other sub- jects who kept caries free over the past 2 years and from the above 10 subjects. The predominant bacteria were isolated by using both the most probable method in media at pH7·0 and 5·2, and the conventional plating methods.Streptococcus spp. was identi- fied by means of API 20 Strep commercial kit.Results S.mutanswas the predominant aciduric bacteria in fissures of caries- free children and more frequently recovered at neutral pH. Whilst,S.sanguiswas predominant at pH5·2 in sound and carious fissures of caries-active children. The proportion of Gram-positive rods at pH5·2 was 82%, significantly greater than that of 61% at pH7·0.Conclusion The ecology of plaque is complex and the composition ofmicroflora at each stage of caries could be differ- ent. The composition of bacteria was consistent with the changes of local pH in the plaque.
    Influences of Applying Force during the Different Stages of Estrous Cycle on Orthodontic Tooth Movement of Rats

    Objective To explore the effects of applying force during the different stages of estrous cycle on orthodontic tooth movement of rats, so as to offer an experimental princinple forwomen′s orthodontic treatment.Methods 80 female 3-month-old Wistar rats, which had a stable and five-day estrous cycle, were used. Theywere randomly divided into control groups and load- ing-force groups. Each group was divided again into 4 sub-groups according to the different stages of the estrous cycle. The load- ing-force groups received repeated intermittent orthodontic force forfourtimes seperately duringthe same stage of the estrous cycle. The distance between upper incisor and the first molar on the left was measured. The data were analyzed by One-way ANOVA, S-N-K.Results Therewere significant variations in the amount of toothmovement amongthe groupswhich received the mechani- cal force duringthe different stages of the estrous cycle (P<0·01). Therewere significant variations in the amount of toothmove- ment between the groups received force during pre-estrus and estrus (P<0·05). There were not significant variations between the groups received force during metestrus and diestrus (P>0·05). The largest amount of tooth movement was in estrus group and smallest in pre-estrus one.Conclusion The effect of tooth movement depended on which stage of estrous cycle was chosen to be the time of applying orthodontic force.

    Objective To reconstruct 3D shape of edentulous jaw by using improved measurement system and to analyze the precision of this method.Methods Standard edentulous jawmodels were measured with a improved measurement system by cut- ting and scanning layer-by-layer, and the 3D shape of the models was reconstructed by image processing with specialty software. Ten cubic plaster modes were reconstructed by the same way, data of every border before and after reconstruction were measured and analyzed.Results Data of edentulous jawmodelswere obtained. The errors in ten cubic before and after reconstructionwere not significantly different(P>0·05), data measured in horizontal plane were not significantly different(P>0·05), and data measured in altitude directionwere significantly different(P<0·01), data measured after reconstructionwere less than before ,the error was (0·09±0·08) mm.Conclusion The improved cutting and scanning measurement system can be fit for the 3D recon- struction of stomatognathic system with high precision.

    Objective To find the reasons why patients always have temporomandibular disorders (TMD) after condylar frac- ture by analyzing the stress distribution change of the condylar surface whose subcondylar fracture ware fixed by miniplate during the whole healing process.Methods Analyzing the stress distribution change of the condylar surface whose subcondylar fracture were fixed by miniplate during the whole healing process by three-dimension finite element method (3D FEM).Results During the whole healing process of the fracture, the miniplate osteosynthesiswas helpful to the biomechanics environment rehabilitation of the condylar local, but it still had differencewith normal afterfixing 12 weeks long.Conclusion The difference of stress distribu- tion of condylar surface that fractured and fixed by miniplate with normal may be part of the reason of TMD after the subcondylar fracture miniplate osteosynthesis.

    Objective To evaluate the psychological situations of cleft lip and/or palate children′s parents, to discuss the rela- tionship between psychological situations and cleft types, and to provide clinical treatment to improve the psychological situations of these parents.Methods 100 parents whose children were proceeded the primary surgery treatment were selected as the study group, and other 34 parents whose children were normal as the control group. All parents were inquired and tested by the life event scale (LES) and self-rating anxiety scale (SAS).Results The LES total scores of the cleft lip and/or palate patients′par- entswere significantly high compared with the normal (P<0·05). The SAS total scores of the cleft lip and/or palate patients′ parents had no significant differencewiththe control group (P>0·05). The scores of cleft lip and/or palate patients′parentswere the highest and the scores of cleft palate patients′parents were the lowest.Conclusion Most of the cleft 1ip and/or palate pa- tients′parents were disturbed by a negative psychological situation. Some kind of negative psychological situation may manifest some body symptoms.Meanwhile,the three subgroups′data had the significant difference,which indicated that parents may much more care about the appearance abnormality.

    Objective To study effective operation method of congenital velopharyngeal insufficiency by reviewing clinical data of 29 patients with pharyngoplasties.Methods 29 patients (9 male and 20 female, ranged from 4 to 25 years old, mean age was 13·5) with congenital velopharyngeal insufficiency were available for this study. All cases received Chinese speech intelligibility test and blowing test before and after the pharyngeal flap surgery.Results Chinese speech intelligibility of 29 cases who under- went pharyngoplasties was improved from 33·86% to 43·13%, and the blowing time was increased from 4·60 to 17·72 seconds, the degree of abnormal speech was reduced.Conclusion The pharyngeal flap surgery is an effective method for congenital velo- pharyngeal insufficiency, which reduces hypernasality and increases oral pressure, improves the velopharyngeal function.

    Objective To investigate the association of estrogen receptor(ER) gene polymorphism and primary trigeminal neu- ralgia.Methods By polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), ER gene polymorphism was analyzed in 20 trigeminal neuralgia(TR) patients and 20 control individuals, and the distribution of ER genotype was com- pared in TR group and control group.Results Therewas no significant difference in frequencies of allele and genotype in XbaI or PvuII polymorphism or XbaI with PvuII polymorphisms together between TR group and control group (P>0·05). The genotypic distribution of Xx or PpXx in TR group was higher than control group, and it was contrary to xx, ppxx or Ppxx in TR group and control group.Conclusion XbaI or PvuII polymorphism may be related toTR. Women with PpXx genotype may be a dangerous factor to primary trigeminal neuralgia.

    Relationship between Periodontal Disease and Rheumatoid Arthritis

    Objective To study a population of rheumatoid arthritis patients and determine the extent of periodontal disease in these patients, in orderto investigate the relationship between periodontal disease and rheumatoid arthritis.Methods The experi- mental group was composed of 70 patients with rheumatoid arthritis and the control group consisted of 70 age- and gender-matched individualswithout rheumatoid arthritis. The relationship between periodontal status in rheumatoid arthritis and control groups as well as the relationship between periodontal status and rheumatological findings in patients were analyzed.Results The percent- age of periodontal disease was statistically significant between experimental and control group (P<0·01). The difference of aver- age number of missing teeth and bleeding on probing in the experimental group and control group were not statistically significant (P>0·05). There were more number of periodontal disease index 5 or 6 in experimental group than in control group (P< 0·05). Rheumatoid arthritis patients with moderate to severe bone loss had deeper degree of morning stiffness, erythrocyte sedi- mentation rate levels and serum C-reactive protein levels than patients with no or mild bone loss.Conclusion Individuals with rheumatoid arthritis are more likelyto experience periodontal disease compares to healthy subjects. They are also very likelyto suf- fer from moderate to severe periodontitis.

    Objective To assess the dental caries prevalence of children in Yinchuan.Methods The 3-, 5-, 12-, 15-year- old children were chosed from nursery, primary and middle schools of Yinchuan in 1990, 1996 and 2001. The surveywas carried out by five senior dentists from stomatologic hospital of Yinchuan according WHO criteria. DMFT, DMFS and the ratio of tooth filled were counted.Results From 1990 to 2001, the prevalence of dental caries of 3-, 5-, 12-year-old children was degression (P<0·01). Butthe prevalence of dental caries of 15-year-old children had no obvious change. The dental caries of small propor- tion of children were serious. The ratio of tooth filling was low.Conclusion With the development of oral health education, the prevalence of dental caries of 3-, 5-, 12-year-old children in Yinchuan is descend. But the diagnosis, prevention and treatment should be strengthened, and the ratio of tooth filling should be increased.
    Carotid Body Tumor: A Retrospective Study of 26 Consecutive Cases

    Objective To study the clinical features, diagnoses and operative intervention of carotid body tumors.Methods  The medical records of 26 patientswith carotid body paragangliomas between 1993 and 2004 were retrospectively reviewed.Results  Two of twenty-six patients had bilateral tumors. One patient had malignanttumorswith cervical lymph nodesmetastases. Twen- ty-one patients were evaluated with a preoperative angiography. No preoperative embolization was performed. Eleven patients un- derwent ultrasonography, nine had CTscan of the neck, five had magnetic resonance imaging. Twenty-six patientswere all operat- ed. Vascular reconstructionwas performed in five cases. The ligation of carotid arterywas carried out in four cases and another pa- tient had vagus nerve resection. No death and recurrence occurred in all cases.Conclusion Angiography and ultrasonography are valuable in the diagnosis of carotid body tumor. Surgical removal should be made early to avoid the possibility of eventual metasta- sis and progressive local invasion. The pattern of operation should be chosed according to the relation of tumor and carotid.
    Investigation on Border Mandibular Movement of Adolescent with AngleⅢMalocclusion in Initial Stage of Permanent Dentition

    Objective To investigate characteristics of the bordermandibularmovement of the adolescentwithAngleⅢmaloc- clusion and to find the correlations between craniofacial morphology and border mandibular movement function.Methods The subjects consisted of 21 adolescent patients with AngleⅢmalocclusion in the initial stage of permanent dentition(mean age 12·3 years). 20 adolescents with individual normal occlusion served as control (mean age 12·8 years). The border mandibular move- ment of all cases in the two groupswere traced and recorded byK6-1 Evaluation System and standardized lateral cephalogramswere taken.Results The results revealed no significant difference in the maximal movement range between two groups, but indicated a positive association between the maximum jawopening and both the mandible length(Go-Gn) and the L1-MP. The distance from ICP to maximal opening (D) was positively correlated with facial height and the L1-MP. While to adolescentswith individual nor- mal occlusion, the wider the maximal jawopeningwas, the bigger the posterior facial height and the cephalometric values of man- dibular ramus. The distance D positively correlated with the anterior lower facial height.Conclusion AngleⅢmalocclusion had its characteristics in the tracing and had a specific relationship with the craniofacial morphology.

    Objective To find an objective indexto describe the severity of hypernasality in cleft palate patients through speech spectroanalysis.Methods 60 audio records of cleft palate patients were analyzed by spectroanalysis, the relative energy in low frequency area (REL) was calculated in the LPC spectrogram. One experienced speech therapist and two nonprofessionals made perceptual judgments of the same records. Spearman correlation analysiswas conducted to test if the 3 groups of data have signifi- cant correlations.Results RELhad significant correlation with professional judgments (correlation coefficient was 0·272,P< 0·05) and nonprofessional judgments (correlation coefficient was 0·274,P<0·05).Conclusion RELcan be used as one ob- jective reference in the judgment of the severity of hypernasality.

    bjective To evaluate treatment outcomes ofmandibular complete denture supported bytwo endosteal magnetic-at- tachment implants in edentulous patients with severe mandibular alveolar ridge absorption.Methods Eight edentulous patients with severe mandibular alveolar ridge absorption were chosen. Each of themwas treated with a complete denture supported by two CDIC implants. The implants were inserted at the areas of two the first premolars respectively. After five months, two magnets were adhered to the inside of the complete dentures.Retention,masticatory efficiency and maximal masticatory force were measured before and after the magnets were adhered and six months later.Results Statistical analysis revealed that retention, masticatory efficiency and maximal masticatory force were all increased after the magnetswere adhered (P<0·01) and sixmonths later(P< 0·01).Conclusion Retention, maximal masticatory force and masticatory efficiency were significantly increased after the en- dosteal magnetic attachment implants were applied. It is concluded that mandibular complete denture supported by two endosteal magnetic attachment implants is an effective method for severe alveolar ridge absorption cases.

    Objective To determine whether dentin matrix proteins were expressed by the human odontoblast-like cell line hTERT-hOd-lin vitro.Methods Collagen typeⅠ, bone sialoprotein (BSP),dentin matrix protein 1 (DMP1) and the marker for odontoblast, dentin sialophosphoprotein (DSPP) and dentin sialoprotein(DSP) were detected in these cells by immunohistoche- mistry, RT-PCR and in situ hybridization. During being cultured in mineralizingmedium for 5 weeks, the secretion of OC and ac- tivity of ALP were measured once aweek.Results DSPP, DMP1,BSP and collagen typeⅠwere expressed in hTERT-hOd-l ei- ther at mRNA or protein level. Underthe induction of mineralizingmedium, the cells showed higher activity of ALP and increased secretion of OC.Conclusion hTERT-hOd-l expressed dentin extracellularmatrixin vitro, which means the cell line has the po- tential of mineralization.

    Objective To evaluate the antibacterial activity of nHA-PA66 on the predominant bacteria of infected root canalin vitro.Methods Agar diffusion method was the testing method.Porphyromonas gingivalis(P.gingivalis),Fusobacteria nuclea- tum(F.nucleatum) andPrevotalla intermedius(P.intermedius) were the tested bacteria. The powder and polymerized film of nHA-PA66 were the testmateialswhile the CCQ and filter paper as the control.Results nHA-PA66′s powder showed good ante- bacterial effect on theP.gingivalisandP.intermedius, its filmwasweaker. Both of them had no effect onF.nucleatum.Con- clusion nHA-PA66′s antibacterial effect was not ideal, for better clinical results, some additives can be included.
    Construction of a Virulence-related Gene Library ofStreptococcus mutansby Suppression Subtractive Hybridization

    Objective To construct a suppression subtractive library of virulence-related genes from c serotypeStreptococcus mutans(S.mutans), and lay foundations for screening the virulent genes.Methods After being isolated from virulent and avir- ulent strain ofS.mutansrespectively, the intact and high-pure genomic DNAwas digestedwith three appropriate four-base-cutting restriction endonucleases to produce fragments of optimal length. The digested DNA of the virulent strain ligated with adaptorwas used as tester DNA, and that of the avirulent strain as driver DNA. Then the suppression subtractive hybridizationwas carried out, and the efficiency of ligation and subtraction detected respectively. The subtracted fragmentswere inserted into vector pCR2.1 us- ingT/A cloning kit, and transformed intoE.coliTOP10F′competent cells. Those white colonies were selected to construct the suppression subtractive library.Results AluⅠchosen from three restriction endonucleases was verified to be suitable for prepar- ing restriction fragments fromS.mutansgenomic DNA. Through electrophoresis ofAluⅠ-digested DNA, a smear ranged from0·1 to 2·0 kbwas observed. The ligation efficiency of tester DNAwith adaptorwas at least higherthan 25 percent. The subtraction ef- ficiency of suppression subtractive hybridization confirmed the success in enrichment of differential genes between virulent and avir- ulent strain ofS.mutans. In the subtracted group, the appearance time of the 23S rRNAgene both in tester and driver DNAwas later than that in the unsubtracted group by six cycles. It suggested that suppression subtractive hybridization happened indeed. After the subtracted fragmentswere cloned, 96 colonieswere picked up for constructing the suppression subtractive library of viru- lence-related genes ofS.mutans.Conclusion Suppression subtractive hybridization allows rapid and easy construction of viru- lence-related gene library ofS.mutans.
    Preparation of Enamel Matrix Proteins Controlled Release Microspheres and Their Biological Effects on the Proliferation and Differentiation of Human Periodontal Ligament Cellsin vitro

    Objective To prepare enamel matrix proteins (EMPs) loaded dextran-based hydrogel microspheres (EMPs-dex- MPs), and to evaluate their EMPs controlled release property and their biological effects on the proliferation and differentiation of human periodontal ligament cells (PDLCs)in vitro.Methods Using dimethylbenzene as the oil phase, EMPs-dex-MPs were achieved by emulsion-chemical crosslinking technique. The process of the recombination preparation was optimized by orthogonal factorization method. The configuration and size of EMPs-dex-MPs were determined by scanning electron microscope. The EMPs loading content and encapsulation rate of EMPs-dex-MPs, and their biodegradation characteristic were studied by routine analysis methods. Dynamic dialysis method was used to determine the release characteristic of EMPs-dex-MPsin vitroand its influencing factors. The proliferation of cultured PDLCswas measured byMTTmethod and the differentiation of PDLCswas measured by their alkaline phosphatase (ALP) activities.Results The results showed that EMPs-dex-MPswere homogenous and stable with the av- erage diameter 25μm, and the EMPs loading content was (32·8±1·2)%, the encapsulation rate was (78·9±1·0)%. Under 9% physiological saline solution contained a very thimbleful quantity of dextranase EMPs-dex-MPs could be biodegraded complete- ly during about 40 days. Theinvitroexperiments showed that about 80% of EMPs could be released out in 20 days. Using EMPs- dex-MPs could enhance the proliferation responses and ALP activities of PDLCs more than 12 days.Conclusion As a new sus- tained release system of growth factors,the dex-MPs is stable, workable and biodegradable. EMPs-dex-MPs, whose drug release can be controlled by preparation technique, may be more effective in promoting periodontal tissue regeneration.
    Construction of the Recombinant Adenovirus Vector Expressing Human CD40 Ligand Gene

    Objective To construct a recombinant adenovirus vector expressing hCD40Lgene and explore it in the use of anti- tumor gene therapy.Methods 1 900 bp gene fragmentwas obtained form plasmid pORF-hCD40L byXhoⅠ/SwaⅠcutting and then cloned directionally into the pShuttle plasmid, finally, the resultant plasmid was digested by restriction endonnucleasePmeI and subsequently cotransformtion into BJ5183 cells with the adenoviral backbone pAdEasy-1 to obtain the homologous recombinant and then the recombinant was packaged in the 293 cells. Some methods such as PCR and endonulease digestionwere employed to identify the recombinant adenovirus.Results The evidences of endonulease digestion and PCR analysis confirmed that recombi- nant hCD40L gene was correctly inserted into adenovirus vector.Conclusion The adenoviral vector which expressed hCD40L gene was constructed. It provides an experimental basis for studies on it expression in the mammalian cells and in tumorgene ther- apy.

    Objective To investigate the adhesion to buccal epithelial cells ofCandida albicansisolates from erosive oral li- chen planus(OLP) and nonerosive OLP, and its role in the development of OLP.Methods A total of 112 isolates, comprising healthy control (26),erosive OLP (62) and nonerosive OLP (24), were screened for the adhesion by using buccal epithelial cell (BEC) assay.Results The adhesion to buccal epithelial cells of the isolates from erosive OLP group was stronger than that of those from healthy control.Conclusion Candida albicans, some isolates with a special virulence attribute may contribute to the occurrence and progression of erosive OLP.
